Effects of the Polypara Ethanol Extract on. Blood Pressure, Blood` Sugar Level, and Transmembrane Potentials of the Cardiac Pacemaker Cells in Experimental Animals.




Abstract
Effects of the ethanol extract of Polypara cordata Buec.(PE) on the blood pressure and the heart rate were investigated in cats. Also explored was the effect of PE on the blood sugar level in the spontaneously diabetic, KK-mouse. On the other hand, the transmembrane potentials of the pacemaker cells of the rabbit heart were registered by conventional microelectrode technique before and after superfusion with the PE.
The results obtained were as follows:
1. At the dose of 300 mg/kg, the PE significantly decreased the arterial pressure and the heart rate in anesthetized cats.
2. Following intravenous administration of 150 mg/kg and 300 mg/kg PE, the car-diac arrest were observed in 20% and 44% of the experimental cases, respectively. The PE-induced cardiac arrest was not affected, by bilateral vagotomy.
3. The blood sugar level -of the KK-mouse was ¡Æsignificantly elevated¢¥ after in traperitoneal administration of the PE (500 mg/kg).
4. Following superfusion with the PE, the action potential amplitude (APA) and the rate of spontaneous firing (RSF) of the sinoatrial node cells were significantly decreased while the action potential duration (APD) was not altered.
5.¢¥When the sinoatrial node cells were superfused with 0.25% or 5.0%, the incidence of electrical asystole was 25% and 64% of the experimental cases, respectively.
6. Electrophysiological effect of the PE on the atrial muscle fiber or the atrioven-tricular node cells were similar to that on the sinoatrial node cells.
